Born on June 16, 1998 in Amagasaki City, Hyogo Prefecture, Japan, Yasutoshi Don played for a professional club (Nishinomiya Junior SS) during his elementary school years, and was invited by Gamba Osaka, Cerezo Osaka, and Nagoya Whale, among other Nippon Globe teams, as he progressed from elementary school to junior high school, and ultimately joined Gamba Osaka's 2011 U-15 ladder team.
In 2015, Don made his official debut in the Gamba Osaka first team. on May 27, 2015, Don made his team debut in Gamba Osaka's AFC Champions League match against FC Seoul, becoming the youngest player in AFC Champions League history to make an appearance at the age of 16 years and 344 days. on June 20, Don made his debut as a debutant in the game against Vigatte Sendai.
In 2016, Gamba Osaka set up a U-23 to officially play in the Japanese C-League, and Yasutoshi Don became the team's starter.

On August 27, 2019, Dutch Eredivisie giants PSV Eindhoven announced on their official website that they have reached an agreement with Groningen on the transfer of Japan international Rui Don. After completing his medical, Rui Don will officially join PSV Eindhoven. According to the German transfer market website, the transfer fee is 7.5 million euros.
PSV Eindhoven has officially announced that Yasutoshi Don has moved to Bielefeld on loan. Don Yasutoshi's loan contract with Bielefeld will expire on June 30 next year.
