In Uruguay's Spanish Central, Besino opened a career chapter that saw him establish himself among the giants of the Uruguayan Nationals with a transfer in 2011.

The wheels of time rolled forward and on August 23, 2013, Besino joined the camp of Serie A side Fiorentina for a transfer fee of €24 million.

The following year, Bessino experienced the ups and downs of a loan spell and was sent to work out at Empoli.

In 2017, Fiorentina released Besino to undergo a medical with Inter, and the transfer deal was finally finalized on August 2, with Besino signing a contract until 2021 with Inter.

With the No. 11 jersey under his belt, he played a full match without dropping a beat against Fiorentina in his debut match on Aug. 20, helping his team to a 3-0 win. He followed that up by contributing a key goal against Roma, helping the team to a 3-1 comeback victory, the first time they had beaten Roma at the Stadio Olimpico in nine years.

On May 20, 2018, Bessino scored the winning goal in a 3-2 win over Lazio to help his team return to the Champions League. On September 18 of the same year, he scored the winning goal in stoppage time against Tottenham Hotspur to help his team win a close 2-1 Champions League match.

On August 1, 2022, Bessino moved to Lazio on a free transfer, signing a three-year contract that marked another important chapter in his career.
